Objective: The increasing demand for more flexible and student-centered educational programs has led to the widespread popularity of E-learning. The success of this approach depends on how E-learning is applied. Considering the challenge of how to teach practical courses electronically, the purpose of this research was to compare the effectiveness of online and offline methods in electronic education on the satisfaction of medical students in the physiology laboratory course. Methods: In this descriptive-analytical study, 112 medical students were divided into two groups. The first group took part in Skyroom classes (online), and the second group participates in Physiology laboratory classes using Navid system (offline) in one academic semester. At the end of the semester, a written evaluation was collected by a validated questionnaire with a three-point Likert scale (complete satisfaction, partial satisfaction, and dissatisfaction). Finally, statistical analysis was performed using SPSS version 21 and one-way analysis of variance. Results: The results of the present study showed that in most of the evaluated items, the level of students' satisfaction with the educational method of using the Skyroom system is significantly higher than that of the Navid system. On the contrary, in factors such as student concentration during teaching, internet productivity and holding exams, the level of students' satisfaction with the educational method of using the Navid system is significantly higher than that of the Skyroom system. Also, there is no significant difference between students' satisfaction with the level of text, audio and image coordination and system security. Conclusion: According to the results of this study, students prefer to participate in an online E-learning program. However, using online classes alone may have limitations. Optimizing suitable internet infrastructure can help to increase the level of satisfaction of students.
